Emerging evidence has suggested that EVI1 regulates transcription, in part, through epigenetic modification.56 For example, the aberrant DNA hypermethylation signature in EVI1-directed acute myeloid leukemia is likely through interaction with DNA methyltransferases 3A and 3B.58 In this study, we observed a significant H3K9me3 enrichment within the promoter regions of VSMC-specific genes (SMαA, SM22α, SRF, and Myocd), and this enrichment was significantly inhibited by EVI1 knockdown (Figure 6F). This evidence concerns the gene SRF and acute myeloid leukemia.
