CASP3 and Hyperglycemia: As expected, Rap attenuated the Bmal1KD-induced enhancement of hyperglycemia cardiotoxicity, as shown by reduced fractions of dead cells in live/dead staining (Bmal1shRNA 36.92 ± 4.88% vs. 26.72 ± 4.10% Bmal1shRNA plus 100 nM Rap, p < 0.05, n = 8; Figure 1A and 1B) and TUNEL-positive cells (29.57 ± 4.30% in Bmal1shRNA vs. 20.59 ± 2.89% in Bmal1shRNA plus 100 nM Rap, p < 0.05, n = 8; Figure 1C–1D) and by lower levels of cleaved caspase 3 and PARP (Figure 1H–1J).
