In the current study, to determine which step of this pathway is involved in growth of ovarian cancer, we first examined the effects on cell growth of several inhibitors of the pathway: 6-fluoromevalonate, which inhibits mevalonate pyrophosphate decarboxylase; YM-53601, which inhibits squalene synthase; Lonafarnib, a farnesyltransferase inhibitor that blocks farnesylation with farnesyl pyrophosphate; and GGTI-298, a geranylgeranyltransferase inhibitor that blocks geranylgeranylation with geranylgeranyl pyrophosphate (Figure 1).
