However, lack of clinical benefit for BACE1 inhibitor verubecestat and for many Aβ-lowering drugs in mild to moderate AD [162,163] suggests that despite successful pharmacodynamic effect in amyloid reduction, a more relevant strategy may be reduction of Aβ production early in the disease process, or as a prophylactic, and is being explored for all BACE1 inhibitors currently in trials ([4,164,165]; see Table 4 in the section below).
