In order to determine the effect of CPS acetylation on immune protection against Burkholderia infection, glycoconjugate vaccines were produced with carrier protein TetHc chemically conjugated by reductive amination [16] to purified B. thailandensis E555:: wbiI (pKnock-KmR) CPS or its deacetylated counterpart (deAc CPS), which other than the absence of the O-acetyl group had exactly the same structure as native CPS (Fig. 1C).
