Aberrant DNA methylation patterns are commonly observed in many cancers.37 UHRF1 plays an essential role in the maintenance of DNA methylation.34 UHRF1 recruits DNA methyltransferase 1 (Dnmt1) to the hemimethylated DNA to ensure efficient maintenance of methylation patterns.34 In RIP3-null RKO cells, knockdown of UHRF1 reduced the methylation level of Rip3 promoter and induced the expression of both RIP3 mRNA and protein, while knockdown of Dnmt1 failed to induce RIP3.
