Single-cell suspensions from tumours, spleens, draining and non-draining lymph nodes were obtained 12 days (the time point previously reported for peak values of antigen-specific tumour-infiltrating lymphocytes (TILs) before tumour rejection5) after tumour cell inoculation and probed simultaneously for all 81 potential T-cell specificities, while staining with 28 different antibodies for the further identification and characterization of CD8+ T cells (Supplementary Fig. 1B and Supplementary Table 2).
