Because Shp2-iSNAP can detect the tyrosine phosphorylation of the BTAM motif from SIRPα, we fused Shp2-iSNAP at the C-tail of SIRPα (SIRPα Shp2-iSNAP) to rewire the anti-phagocytic ‘don’t eat me’ CD47-SIRPα-Shp1-negative signaling8, 17 into a positive Shp2 action17, 21 upon the engagement of SIRPα by CD47, thus facilitating phagocytosis of opsonized tumor cells initiated by IgG-FcγR interactions (Fig. 2a).
