NLRP3 and age-related macular degeneration: The most notable pathogenic processes in AMD include (1) the accumulation of discrete drusen deposits (composed of partially degraded oxidized products) between the RPE and Bruch membrane (BM), (2) intracellular accumulation of lipofuscin granules in the RPE and its constituent bis-retinoid N-retinylidene-N-retinylethanolamine (A2E), (3) chronic local inflammation, (4) innate immune activation of the complement system by the inflammasome nod-like receptor-P3 (NLRP3), (5) mtDNA damage, and (6) oxidative and endoplasmic reticulum stress.
