An impairment of this process in the PINK1-dependent mitophagy pathway through the loss of PGAM5 causes an accumulation of damaged mitochondria that worsen necroptosis, dopaminergic neuron degeneration, and defects in growth and survival, establishing a molecular link between PGAM5 and the pathogenesis of Parkinson's and cardiac diseases (Brenner et al., 2013, Imai et al., 2010, Kanamaru et al., 2012, Lu et al., 2014, Lu et al., 2016, Remijsen et al., 2014, Wai et al., 2016).
