Indeed, we have shown previously that disruption of the Disc1 gene in mice results in altered expression of Nrxn1,33 a gene in which copy number variation, common variants and rare variants are associated with schizophrenia.60, 61, 62 The hypothesis of a distal effect of variants in the Interactome on disease risk is also consistent with the recent association of copy number variants linked to intellectual disability with schizophrenia in a much larger sample.63 The gene discussed is DISC1; the disease is schizophrenia.
