Given the observed EMT in K-RasV12/αV-RFP cells, we focused our analysis on the known EMT-inducing transcription factor genes SNAI1, SNAI2, ZEB1, ZEB2, TWIST1 and TWIST2, all of which have been implicated in cancer cell metastasis.40 Although RNA-Seq analysis identified only ZEB1 as a gene highly upregulated by K-RasV12/αV-RFP co-expression, a qPCR analysis detected more than 395-fold upregulation of TWIST1 and 5.5-fold upregulation of SNAI2 (Figure 7c). The gene discussed is TWIST2; the disease is cancer.
