In a proof-of-mechanism study from a French group [81], 20 patients with chemotherapy-naïve primary or relapsed WDLS/DDLS, MDM2 amplified and eligible for resection, received RG7112, a MDM2-antagonist, with one response and 14 stable disease cases being reported, albeit with a significant gastrointestinal and bone-marrow toxicity. Here, MDM2 is linked to well-differentiated liposarcoma.
