Among the top 20 genes in the MPNST vs. NF signature are 4 genes reported as induced in the transition from benign NF to MPNST (S11 Table); their products are the topoisomerase TOP2A, needed for correct chromosome segregation in mitosis, the apoptosis inhibitor BIRC5, a member of the mitotic chromosome passenger complex, the architectural transcription regulator HMGA2, and TPX2, which is essential for correct mitotic spindle assembly and activates the AURKA kinase to control cell cycle progression. The gene discussed is HMGA2; the disease is malignant peripheral nerve sheath tumor.
