Studies using PTH analogs in vivo have suggested that selective stimulation of the endosome-mediated long signal and the prolonged half-life of PTH in the blood are favorable for inducing hypercalcemia.10,11 However, the association between the temporal variations in PTH-mediated cAMP signaling and the downstream cellular responses that promote bone anabolism and catabolism remains unclear.
