ALK and non-small cell lung carcinoma: Ceritinib (Novartis) is 20 times more potent than crizotinib and was developed in the clinic in a small, biomarker-driven phase I study of ALK-translocated NSCLC, in which 66% of patients were previously treated with crizotinib, demonstrating excellent response rates of 58% and a PFS of 7 months (35), leading to FDA approval of the drug after a phase I study (a first in the modern oncology era).
