As shown in Figure 3A, we constructed the luciferase reporter plasmids containing the wide type 3′UTR of notch3 or the mutated 3′UTR of notch3, and pancreatic cancer cells transfected with miR-613 mimics and reporter plasmids containing the wide type 3′UTR of notch3 significantly suppressed the luciferase activity when compared to cells transfected with scrambled miRNA and reporter plasmids containing the wild type 3′UTR of notch3 (Figure 3B). The gene discussed is NOTCH3; the disease is familial pancreatic carcinoma.
