Interestingly, in addition to their well-characterized role in cancer, postzygotic somatic mutations in PIK3CA have also recently been identified in a spectrum of overgrowth disorders including congenital lipomatous overgrowth with vascular, epidermal, and skeletal anomalies (CLOVES) syndrome, PIK3CA-related overgrowth spectrum (PROS), Proteus syndrome (PS) and other AKT-related disorders. Here, AKT1 is linked to Proteus syndrome.
