Regarding the conversion from MCI to LOAD, one study found that participants harbouring 6 or more AD risk alleles progressed to AD twofold (HR 1.89 [95% CI 1.01–3.56]) more rapidly than those with only 6 alleles [12], whereas researchers in a second study observed that an AD GRS composed of 19 loci was associated with the conversion to dementia (HR 1.59 [95% CI 1.23–2.05]), but only when apolipoprotein E (APOE) was included in the risk score [9].
