Second, in rescue experiments, whereby cells were transfected with control or ALPK1 siRNA and then transfected with the empty vector pEYFP or a siRNA-resistant full-length ALPK1-YFP cDNA construct (Fig 6I and 6J), overexpression of YFP-ALPK1 did not induce the formation of TIFA oligomers in the absence of infection, indicating that this process was tightly regulated.
