Several high‐risk mutations have been identified in melanoma‐dense families, including mutations in the cyclin‐dependent kinase inhibitor 2A (CDKN2A) gene (Cannon‐Albright et al., 1992), the cyclin‐dependent kinase 4 (CDK4) gene (Zuo et al., 1996) and more recently in the Breast cancer 1 (BRCA1)‐associated protein 1 (BAP1) (Aoude et al., 2013; Wiesner et al., 2011) and protection of telomeres 1 (POT1) genes (Robles‐Espinoza et al., 2014; Shi et al., 2014). The gene discussed is POT1; the disease is melanoma.
