The conclusion that BAF180 protein's tumor-suppressive activity in ccRCC is HIF1 dependent is also supported by functional studies, in which expression of HIF1α in H2 786-O cells switches BAF180 from a oncogenic protein to tumor-suppressive protein (Figures 4b and c), although knockdown of HIF1α in H1H2 SLR25 cells turns BAF180 from a tumor-suppressive protein to a tumor-promoting protein (Figures 4e and f). This evidence concerns the gene PBRM1 and nonpapillary renal cell carcinoma.
