No DFNA5 or GSDMD processing was observed in S100 lysates from the caspase-3-deficient MCF7 breast cancer cell line26, indicating that caspase-3 is the primary protease responsible for processing of DFNA5 and GSDMD downstream of the apoptosome (Supplementary Fig. 3b, left panels, and Supplementary Fig. 3c). The gene discussed is CASP3; the disease is breast carcinoma.
