Further supporting this clinical observation, knockdown experiments involving the treatment of GC cells with STAT3 siRNA in the presence of DZNep demonstrated an increased apoptotic rate, as well as enhanced caspase-3/9 activity (Figs. 3e and f, P = 0.003, P = 0.027, respectively), which in turn resulted in the down-regulation of EZH2 at both mRNA and protein levels.
