MAPT and amyloidosis: The amyloid-cascade hypothesis posits that the unregulated synthesis of Aβ and the concomitant formation of extracellular amyloid plaques leads to intracellular neuron injury that is mediated by the formation of tangles of phosphorylated tau protein (O’Brien and Wong, 2011; Revett et al., 2013; Hampel et al., 2015).
