We hypothesized that the combination of 2DG and TRAIL would be highly specific for cancer cells not only because of the preferential uptake of 2DG, but also because cancer cells tend to express much higher levels of DR4 and DR5 relative to normal cells.22 Upregulation of DR5 surface expression with 2DG treatment was found in all cell lines tested; however, abrogation of DR5 expression was not sufficient to eliminate the synergy between 2DG and TRAIL, suggesting its upregulation was not the only determinant of TRAIL sensitivity conferred by 2DG treatment.
