Based on the promising results from preclinical trials [42], the encouraging activity reported in early clinical trials enrolling gastric cancer patients [43], and the great efficacy of the combination of trastuzumab and pertuzumab in breast cancer [44], the phase III JACOB trial (BO 25114) was designed to assess whether the addition of pertuzumab to cisplatin, 5-Fluorouracil and trastuzumab may further improve the survival of HER2- positive gastric cancer patients with an acceptable safety profile [45].
