The arsenal of oncolytic adenoviruses as monotherapy in clinical trials is completed by the so called Delta-24-RGD Adenovirus (DNX-2401), which exploits Rb/p16 deficiencies in cancer cells for replication processes.50 DNX-2401 entered clinical trials as a monotherapeutic particle in a phase 1 trial on brain-cancer patients, administered as single-shot therapy directly into the tumor (NCT00805376; see Supplementary Figure S5c). This evidence concerns the gene RB1 and cancer.
