FADD and neoplasm: As a cytotoxic cytokine, TRAIL selectively induces apoptosis in tumor cells through homotrimeric binding to the membrane-bound death receptor 4 (DR4, also known as TRAIL-receptor 1 or TNF receptor superfamily member 10A) and death receptor 5 (DR5, also known as TRAIL-receptor 2 or TNF receptor superfamily member 10B), the recruitment of Fas-associated death domain protein (FADD), the formation of the death-inducing signaling complex (DISC), and the subsequent activation of caspase-8 and effector caspase-3 (extrinsic pathway).
