Targeting mTOR has demonstrated anti tumouractivity in clinical studies; as a result, allosteric inhibitors of mTOR, have beenapproved for the treatment of renal cell carcinoma, pancreatic neuroendocrinetumours and advanced hormone receptor positive, HER2-negative breast cancer(Hudes et al, 2007; Motzer et al, 2008; Yao et al,2011; Baselga et al, 2012).
