Additionally, the overexpression of a mutant PLA2G16 construct in osteosarcoma cells appears to have dominant-negative activity over the wild-type enzyme as the endogenous levels of p-Erk are reduced (Figure 6) and the xenograft model also shows reduced tumor growth in cells expressing the mutant PLA2G16 compared to empty vector controls (Figure 8B, 8C).
