Using 2-DG and oligomycin to block glycolysis and oxidative phosphorylation generated ATP, respectively, we found that COUP-TFII knockdown was still able to reduce ATP levels in oligomycin treated cells, but not in 2-DG treated cells (Figure 3E), suggesting that COUP-TFII mainly regulates ATP generated by glycolysis in prostate cancer cells.
