Since MLN8237 showed single-agent activity in several neuroblastoma mouse xenograft models during experiments conducted by the Pediatric Preclinical Testing Program, leading to testing in pediatric neuroblastoma patients (Maris et al., 2010), and MLN8237 was more potent at a lower dose than the dual Aurora inhibitor VX680 when combined with ABT-199 (Figure 4C), we decided to pursue ABT-199/MLN8237 combinations in MYCN-amplified neuroblastoma cells.
