To the best of our knowledge, this is the first article that (a) presents genome-wide significant SNPs associated with epigenetic age acceleration, (b) elucidates the underlying mechanism using cis-eQTL studies, (c) shows that the expression levels of one of the implicated genes (MLST8) increase with chronological age, (d) shows that epigenetic age relates to a subunit (MLST8) of both mTOR complexes and (e) shows a significant overlap between genes related to the epigenetic age of nervous tissue (cerebellum) and those implicated in AMD, Alzheimer's disease and Parkinson's disease. This evidence concerns the gene MTOR and early-onset autosomal dominant Alzheimer disease.
