To further confirm that SPAG9 is a functional target of miR-141, we transfected miR-141 overexpressing HCC cells with SPAG9 plasmid, which encrypted the full-length coding sequence of SPAG9 without its 3′-UTR and found that the overexpression of SPAG9 lacking its 3′-UTR substantially reversed the tumor-suppressive effects of miR-141 (Fig. 3).
