Specifically, germline mutations in mismatch repair (MMR) genes MLH1, MSH2, MSH6 and PMS21, and in DNA polymerases POLD1 and POLE2 predispose to a high incidence (lifetime risk 30–71%2, 3, 4, 5) of both CRC and EC. This evidence concerns the gene POLD1 and colorectal carcinoma.
