As our in vitro study reveals the importance of SIRT1 in HSF1 protein stability, we observed reduced SIRT1 protein levels in the midbrain area of both α-syn transgenic mice and the inferior parietal lobes of human patients with DLB (Supplementary Material, Fig. S5A and B), all of which were previously identified to be lack of HSF1 protein (Figs 3 and 4), although the midbrain of the AAV-α-syn injected mice did not show any change in SIRT1 expression levels (Supplementary Material, Fig. S5C). This evidence concerns the gene HSF1 and Lewy body dementia.
