TRz is a humanized monoclonal antibody that binds to Her2 and is used clinically either as an unconjugated antibody (“Herceptin”)38 or as the ADC TRz-emtansine (“Kadcyla”).39 Her2 is overexpressed in 15–20% of diagnosed breast cancers,40,41 where it forms oncogenic signaling dimers with other, ligand activated, receptors from the ErbB family.
