A nonsynonymous sequence variant (rs738409) of the human patatin-like phospholipase domain containing gene 3 (PNPLA3) resulting in the amino acid substitution p.Ile148Met has been identified as major determinant of increased liver fat content and key susceptibility factor for NAFLD in adults [5]. This evidence concerns the gene PNPLA3 and metabolic dysfunction-associated steatotic liver disease.
