Compared with PKM2(WT)-expressing PARP14-depleted HCC cells, PKM2(T365A)-expressing PARP14-depleted HCC cells exhibited reduced levels of PKM2 activity that correlated with a significant increase of GSH levels and decrease in apoptosis (Fig. 9d), supporting a role for PKM2 Thr365 phosphorylation in regulating PARP14-mediated HCC cell survival.
