If, as conjectured here, a neurotoxic phenotype occurs in the aged and AD brain because of co-stimulation by Aβ and other TLR4 ligands (such as LPS), then the model suggests that drug combinations that include a TLR4 blocker, such as the non-opioid form of naloxone, would help to reduce that neurotoxicity (opioid forms that block both TLR2 and TLR4 might help even more).
