Here, we observed elevated expression of gankyrin in clinical cirrhosis biopsies (Supplementary Figure S1), consistent with our previous studies that gankyrin gradually increased during DEN-induced liver cirrhosis and HCC in rat.13 Increased expression of gankyrin in human HCC correlated with poor survival and disease recurrence after surgery.11 To determine the contribution of gankyrin to hepatocarcinogenesis in vivo, we constructed a new transgenic mouse strain gankyrinhep, in which a human gankyrin protein expresses specifically in the hepatocytes driven by the albumin promoter.
