Postzygotic mutations in the PIK3CA gene have also been identified in distinct overgrowth syndromes such as CLOVES (congenital lipomatous asymmetric overgrowth of the trunk, lymphatic, capillary, venous, and combined-type vascular malformations, epidermal nevi, skeletal and spinal anomalies), HHML (Hemihyperplasia Multiple Lipomatosis) and fibroadipose overgrowth (FAO) [4–6]. The gene discussed is PIK3CA; the disease is vascular malformation.
