Oncogenic BRAF (but not wildtype BRAF), which is mutated in up to 50% of melanomas, also regulates MITF via simultaneously stimulating MITF activation through ERK phosphorylation, which leads to its subsequent degradation, and by inducing transcription of MITF via BRN2 upregulation (Davies et al., 2002; Wellbrock et al., 2008).
