PDK3 and Charcot-Marie-Tooth disease: Recently, a mutation conferring PDK3 hyperactivity has been reported as a cause of Charcot-Marie-Tooth disease (Kennerson et al 2013) and by inhibition of PDK3 activity, phenylbutyrate may prevent PDHC hyper-phosphorylation that has been recognized as the underlying cause of peripheral neuropathy.
