A second-generation investigational AURKA inhibitor (MLN8237/alisertib, Millennium), which has 200-fold selectivity for AURKA over AURKB (aurora kinase B) in cell assays, has been recently investigated as monotherapy in phase I clinical trials in patients with advanced solid tumors or lymphomas (NCT01898078, NCT00962091).
