IGF1R and neoplasm: Although available drugs (apart from AZ4547) inhibit more than just the FGFR system, preclinical work confirms that pure inhibitors of FGFR 1 will inhibit growth of FGFR1 amplified tumor cell lines; IGF1R (insulin-like growth factor receptor) is overexpressed in some SQCCs, acting via the canonical growth and survival pathways, but promising phase II results in SQCC lung were not replicated in two phase III trials of figitumumab (a monoclonal antibody against IGF1R).
