EGFR and breast cancer: This illustrates an example of the therapeutic advantage of co-treatment with the benefit of reduced toxicity of conventional drugs.24 Previously, RNAi-mediated inhibition of IAPs in combination with ErbB antagonists, such as the EGFR inhibitor gefitinib, profoundly enhanced apoptosis in breast cancer cells.25 However, although some instances of single small molecules affecting multiple targets have been reported, none have been shown to inhibit EGFR and IAPs.26
