PC-3 (a castration-resistant cell line) showed higher EFEMP1 re-expression upon DAC treatment compared with the other two PCa cell lines (although it showed the higher methylation levels), suggesting that DNA methylation might be more relevant for ‘locking’ gene silencing in more clinically advanced PCa, whereas histone PTMs, a more dynamic transcription regulator, might be more relevant in the early stage, androgen-responsive, tumours.
