It is noteworthy that our data demonstrated for the first time that TAp73 and ΔNp73 are regulated by Nrf-2 through a regulatory region in the different promoters of p73 gene, and treatment with 5-aza-dC can enhance Nrf-2 to bind to the P1 promoter and inhibit Nrf-2 to bind to the P2 promoter resulting in the regulation of p73 expression, which was confirmed in vitro and vivo. To the best of our knowledge, this is the first report revealing that the up-regulation of TAp73 and down-regulation of ΔNp73 induced by 5-aza-dC are correlated with Nrf-2 expression in breast cancer cells.
